Folate Receptor 1 (FOLR1, also FRα/FBP/FOLR) is a GPI-anchored cell-surface glycoprotein and a clinically validated promising target for solid tumors. This protein binds folate with high affinity and mediates intracellular folate transport to meet the demand for one-carbon metabolism required for tumor cell proliferation. FRα is highly expressed in multiple solid tumors including ovarian cancer, endometrial cancer, non-small cell lung cancer and triple-negative breast cancer, while exhibiting very low expression in normal tissues and is spatially inaccessible to circulating molecules, leading to low off-target risks. It serves as a core target for the development of ADCs, therapeutic antibodies and folate-drug conjugates, and FRα-targeted ADCs have achieved marketing approval.
