4-1BB (CD137/TNFRSF9/ILA) is a co-stimulatory receptor of the TNF receptor superfamily. Expressed mainly on activated T cells with higher levels in CD8+ than CD4+ T cells, it is also detected in dendritic cells, NK cells, granulocytes and vascular wall cells under inflammation. CD137 crosslinking promotes T-cell proliferation, IL-2 secretion, survival and cytolytic function. The 4-1BB/4-1BBL pathway boosts secondary CTL responses against viruses and anti-tumor immunity in mice, although it also enhances TCR/CD3-triggered activation-induced T-cell apoptosis. CD137 interacts with TRAF2 for downstream signaling, making it a promising target for human cancer immunotherapy
