ManualCOAMSDSKallikrein-2 (KLK2), a secreted serine protease of the peptidase S1-kallikrein subfamily, harbors a single peptidase S1 domain and is abundantly expressed in the human prostate. KLK2 cleaves Met-Lys and Arg-Ser bonds in kininogen to produce Lys-bradykinin, with preferential proteolysis of Arg-|-Xaa bonds in small-molecule substrates. Its selective release of kallidin (lysyl-bradykinin) from kininogen occurs via hydrolysis at Met-|-Xaa or Leu-|-Xaa positions. Serpins such as protein C inhibitor, antichymotrypsin and plasminogen inhibit KLK2. KLK2 is established as a biomarker for prostate cancer.
