Guanylyl cyclase C (GUCY2C, heat-stable enterotoxin receptor), a type I transmembrane guanylate cyclase, transduces signals via cGMP generation. The GUCY2C paracrine axis, comprising GUCY2C and its ligands guanylin/uroguanylin, protects intestinal mucosa from mutagenic, chemical and inflammatory damage. Murine GUCY2C-targeted CAR-T cells specifically recognize and eliminate GUCY2C-positive human colorectal cancer cells. This work establishes a human GUCY2C-specific CAR-T therapeutic strategy for metastatic colorectal cancer with GUCY2C expression.
